Aim: Despite its antineoplastic effect, doxorubicin (DOX) causes significant oxidative stress and cellular damage in the liver. This study aimed to demonstrate the oxidative damage induced by DOX in liver tissue at the biochemical, histological, and molecular levels and to evaluate the protective effects of Montelukast (ML) on this damage. Material Method: To further evaluate the oxidative stress induced by DOX in liver tissue, superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) activities, reduced glutathione (GSH) levels, and malondialdehyde (MDA) levels were analyzed (p<0.05). Results: Histopathologically, portal congestion, pyknotic nuclei in hepatocytes, cellular degeneration, and sinusoidal dilation were observed. In the DOX + ML group, oxidative stress markers improved: SOD, CAT, GPx, and GSH increased, while MDA levels decreased; histological findings were attenuated, and apoptotic gene expression (Bax, Bcl -2, Caspase -3) was significantly modulated compared to the DOX group. Conclusion: The findings suggest that ML provides a significant hepatoprotective effect against DOX -induced oxidative liver damage.
