Introduction: Despite its widespread therapeutic use, methotrexate (MTX), an antimetabolite agent, is associated with significant side effects. Therefore, this study aimed to investigate the protective properties of ellagic acid (EA) against MTX-induced nephrotoxicity in a rat model. Materials and Methods: Thirty-two Wistar rats were randomly assigned to four groups: a control group receiving saline containing 10% dimethyl sulfoxide (DMSO), an EA group receiving 75 mg/kg EA intragastrically, an MTX group receiving a single 20 mg/kg dose of MTX intraperitoneally, and an MTX + EA group receiving a single dose of MTX on the first day and 75 mg/kg EA intragastrically once daily for 10 consecutive days. Results: Compared with the control group, MTX administration resulted in marked functional impairment and histopathological damage in renal tissue. EA treatment partially reduced MTX-induced oxidative stress, renal dysfunction, and histopathological changes compared with the MTX group. Conclusion: The findings of this study suggest that EA may help reduce MTX-induced kidney injury and may have potential as a supportive agent against MTX-related nephrotoxicity.
